18 families in which three and more relatives suffered from nonpolyposis colorectal cancer have been selected from the 5,500 pedigrees entered in Moscow Cancer Family Register. 9 families met the Amsterdam criteria of Hereditary Nonpolyposis Colorectal Cancer (HNPCC) while in another 9 families (designated as the Familial Colorectal Cancer) the clear-cut HNPCC according to the above mentioned criteria have not been not revealed. In the previous study the hereditary mutations were revealed in four of HNPCC families: hMLH1 in one family and hMSH2 in three other. These four families were designated as HNPCCmut+ while another 5 families were designated as HNPCCmut?. The purpose of the present study was to establish the clinical and genealogical differences between three types of families. HNPCCmut+ group has been shown to differ from another group by relatively high incidence of endometrium and ovary cancer as well as breast cancer. Primary multiple cancers could be detected in half of all female cancer patients belonging to this group. The incidence of stomach cancer among women of both Hereditary and Familial groups was higher then expected proceeding from the population incidence rate.
